Methods described in  Amsterdam and Hopkins 2006 and Jao et al. 2008 The viral construct contains the core viral components, e.g. long terminal repeats (LTRs) and a packaging signal and the gene activation cassette, which includes a polyadenylation signal (pA) from bovine growth hormone, fourteen tandemly repeated UAS14 sequences, and a synthetic element that includes a splice donor (SD).
