Phenotypic analysis of tyrb;csf1ra;pnp4a triple mutants at different developmental stages. Whole fish images of wild-type fish (A, E, I, M, Q) and amber mutants (C, G, K, O, S) at different developmental stages. Eye images of wild-type fish (B, F, J, N, R) and amber mutants (D, H, L, P, T) at different developmental stages. (A-B) At 6 dpf, melanophores, xanthophores and iridophores were observed on the skin and iris of wild-type fish. (C-D) The body of amber mutants was colorless and transparent due to the loss of visible melanophores, xanthophores and iridophores. (E-F) At 10 dpf, the skin of wild-type fish was covered by a large number of pigment cells, and the body was opaque. (G-H) The body of amber mutants remained colorless and transparent, with their internal organs clearly visible. (I-J) At 20 dpf, the body of wild-type fish was completely covered by pigment cells, and was forming vertical bars. (K-L) The body of amber mutants remained colorless and transparent, with their internal organs clearly visible, a small number of iridophores were recovered on the iris. (M-P) At 40 dpf, compared with wild-type fish, the amber mutants remained colorless and transparent, with increasing iridophores recovered. (Q-T) At 180 dpf, the color pattern of wild-type fish is completely stable. Both wild-type fish and mutants were covered with iridophores on the body surface (Q, S) and iris (R, T) due to the recovery of iridophores in mutants, and the body was no longer transparent. (U-X) Some close-ups of amber mutants were taken at 6 dpf, 10 dpf, 20 dpf, and 40 dpf, respectively. Red arrowheads indicate internal organs.
