miR-183 cluster expression in WT and KO TL strain zebrafish retinas. (A,B) RNAscope results with probes specifically binding to miR-183, 96, and 182. Neither the dorsal (A) nor ventral (B) side of the mutant retina show binding with the miR-183 probe. The miR-96 probe binds the retina normally while miR-182 shows decreased binding in the miR-183 KO retina. Magenta indicates probe binding to the retina. The darker the color, the higher the miRNA expression level. Blue indicates nuclei and brown is intrinsic pigment of the RPE. (C,D) Statistical results of RNAscope signal intensity of the above images, the values are the comparison of each miRNA with WT. The results show that miR-183 expression is completely knocked out in this zebrafish model and that miR-182 expression is partially reduced. WTTL: wild-type zebrafish of a TL background, N = 6. miR-183−/−: miR183 11 bp deletion including whole seed sequence, N = 10. Bars in the graph represent s.e.m. ** p < 0.01, *** p < 0.001.
