Generation of the miR-183−/− zebrafish model. (A) Conservation of mature and seed sequences of miR-183 in mammals. (B) Conservation of mature and seed sequences of miR-183 in non-mammals. The mature sequence of miR-183 is slightly different in non-mammals, but the seed sequence is highly conserved. (C) Transcription activator-like effector nuclease (TALEN) technology was used to generate the miR-183−/− zebrafish model. The left and right recognition sequences of TALEN were designed to flank and delete the seed sequence. The NLS sequence helps guide TALEN into the cell nucleus and target the genome, improving editing efficiency. (D) Genotyping of miR-183 knockout zebrafish. A total of 11 bp, including the seed sequence, was deleted. NLS: nuclear localization domain. RVD: repeat-variable di-residue.
