Knockdown efficiency and specificity of Llgl1 morpholinos. (A) Western blot of 36 hpf embryo head extracts probed with anti-Llgl1 antisera (Dollar et al., 2005). Bands vary between 100 and 120 kDa, at the predicted sizes of Llgl1 phosphorylation isoforms. γ-Tubulin was probed as a loading control. (B) Percentage of embryos that showed a wild-type phenotype under llgl1 UTR morpholino only and llgl1 full-length mRNA co-injection conditions. Note the dose-dependent rescue of llgl1 morphants to wild-type phenotype with co-injection of llgl1 mRNA. |
