Expression of the stm-l gene during embryonic development. Dorsal (A-C,E-H,J), ventral (K), and lateral (D,I) views of embryos. stm-l gene exhibits a specific expression in the otic structures (A-D) and nasal placode (E-H, arrows). The stm-l expression in the mouth region is detectable from 6 days postfertilization (dpf; G, arrowheads). stm-l is also expressed in the spinal cord (I), caudal intestine (I), epiphysis (J), and gills (K). Pax2/stm-l interaction is involved in otolith formation. A: Binding ability of zebrafish Pax2 on the upstream region 1 of stm-l (for the sequence see Table 1) in presence of increasing concentrations of unlabeled competitor. B: Dorsal views of medaka embryos at stage 24. Misexpression of zebrafish pax2 leads to ectopic expression of stm-l gene close to the otic region (arrows). C: Lateral views of embryos at 4 days postfertilization (dpf). Misexpression of pax2 or PD-Vp16 (a transcription activation form of Pax2/5/8 protein family) constructs lead to ectopic formation of otoliths (arrows). Heat shocks were done before otoliths begin to form (stage 22). Numbers 1 and 2 indicate the endogenous otoliths. |
