⍺TAT1 is not required for zebrafish development (A) Schematic of the 305-amino acid ⍺TAT1 protein showing the N-terminal catalytic domain. The atat1sa9581 allele introduces a premature stop codon and the atat1b1514 allele contains a frameshifting insertion in exon 1. Both alleles truncate the protein upstream of the catalytic domain. (B) Immunofluorescence of 5 dpf wild-type sibling and atat1 mutant larvae stained for ⍺-tubK40Ac and total ⍺-tub. Scale bar, 1 mm. (C) Lateral views of wild-type sibling and atat1 mutant embryos at 48 hpf (left) and 3 mpf (right). Scale bar, 0.5 mm and 1 cm, respectively. (D) Wild-type sibling and atat1 mutant embryos injected with MO⍺TAT1. Asterisk indicates oedema; arrows indicate body curvature (left). Percentage of embryos exhibiting malformations is quantified (right). Dots represent individual clutches; error bars indicate standard deviation. P values were calculated using the Mann-Whitney U test. Scale bar, 0.5 mm. |
