Molecular features of the ENS in ret mutant zebrafish

(A) Uniform manifold approximation and projection (UMAP) analysis of enteric nervous system (ENS) cells shows all 11 subclusters. UMAP atlas was separated between wildtype and ret mutant zebrafish.

(B) Feature dot plot represents the expression level of the top 5 transcripts (ordered by average log2 fold change) per subcluster of ENS between wildtype and ret mutant zebrafish samples. The colored dot on the top indicates subclusters, as shown in (A), and features were separated according to subclusters.

(C) Expression pattern of ret in the UMAP atlas of ENS, separating between wildtype and ret mutant zebrafish samples. Navy-blue indicates lower expression levels, while yellow indicates higher expression levels. Gray shows cells with no ret expression.

(D) Cluster size comparison of each ENS subclusters between wildtype and ret mutant zebrafish cells. Significantly differentially distributed subclusters are represented in orange. The significance of differences was determined by a threshold of the false discovery rate (FDR) < 0.05 and an absolute log2 fold change >0.58. Dotted line indicates the threshold of log2 fold change.

(E) Bar plot (left) and stacked bar plot (right) show the subcluster size number of the ENS that contributes to each subcluster of all ENS in wildtype (light-blue) and ret mutant zebrafish (light-purple) samples.
