Comparison of SLC45A2 proteins from different species and molecular modeling of the SLC45A2(W74R) variant. (a) Evolutionary conservation of tryptophan (W) at position 74, from fruit fly to human. The W74R mutation changes hydrophobic tryptophan to positively charged, hydrophilic arginine. (b) Molecular modeling using the AlphaFold 3 Server and the protein structure visualization tool UCSF ChimeraX. The W74R mutation introduces a physically impossible overlap with Tyr70, indicating severe structural destabilization.
