Neural crest cell-derived craniofacial cartilage is reduced in polr3asa907/sa907 zebrafish. (A-F) Examination of the craniofacial cartilage using sox10:egfp (green) and all nuclei (DAPI, blue) shows a reduction in overall head size and cartilage elements at 5 dpf and 4 dpf, including Meckel’s cartilage (M) and the ceratobranchials (cb). No changes were detected at 3 dpf and mutants appear phenotypically normal. 3 dpf: n = 12 controls, 12 mutants; 4 dpf: n = 12 controls 12 mutants; 5 dpf: n = 17 controls, 16 mutants. (G,H) Cartilage was examined at 5 dpf using sox9a:egfp. A reduction in size is evident for in the basihyal (bh), while Meckel’s cartilage (M) shows a change in morphology. ch, ceratohyal. (I) Quantification shows that the overall size of Meckel’s is reduced (*p = 0.035). (J) Cell number within Meckel’s cartilage was unchanged (ns, p = 0.367). n = 18 controls, 13 mutants.
