Cellular expression patterns of six genes that were reduced in the GCL and/or INL by Irx7 knockdown at 52 hpf. The expression of six genes, including (A) barhl2, (B) ckmt1, (C) irx4a, (D) pbx1a, (E) robo2 and (F) tfap2a, was reduced in the GCL and/or INL by Irx7 knockdown. These genes all expressed at a lower level in the Irx7-morphant retinas compared with the controls; indicating that they might be normally activated by irx7. The dorsal-tilted and ventral views of the eye are shown for the whole-mount samples; whereas the ventral view is shown for the dissected eyes. Cellular expression patterns of five genes that were reduced in the ONL by Irx7 knockdown at 52 hpf. The expression of five genes, including (A) gnat1, (B) nme2l, (C) aanat2, (D) ndrg1a and (E) rcv1, was reduced in the ONL by Irx7 knockdown. These genes all showed a lower expression level in the Irx7-morphant retinas compared with the controls; thus, they might be normally activated by irx7. For (A) gnat1 and (B) nme2l, the lateral and ventral views of the eye are shown; while for (C) aanat2, (D) ndrg1a and (E) rcv1, the dorsal-tilted and ventral views of the eye region of whole-mount samples, as well as the ventral view of the dissected eyes are shown. The specific expression locations of these genes are listed in Table 2. For (D) ndrg1a, even though there seemed to be expression in the ONL in the whole-mount images (while arrows); the dissected eyes indicated the expression was only detected in the ventral patch (red arrows). Bhlhe22 was over-expressed and cdh11 was not affected by Irx7 knockdown at 52 hpf. The expression level of (A) bhlhe22 in the Irx7-morphant retinas was higher than the controls at 52 hpf, suggesting that they might be normally suppressed by irx7. (B) The expression of cdh11 was not detected in the retinas in either the control or Irx7 morphants. The dorsal-tilted and ventral views of retina are shown. The specific expression locations of these genes are listed in Table 2. Scale bar = 50 μm.
