Activation of an amino acid for incorporation into a peptide by a nonribosomal process, catalyzed by subunits of nonribosomal peptide synthase. The amino acid is adenylated at its carboxylate group (ATP-dependent) then transferred to the thiol group of an enzyme-bound phosphopantetheine cofactor. (2)
