Inhibition of the AP-1 pathway does not recover thyroid function in Nrf2−/−-derived thyroid organoids.
(A) Gene expression analysis shows that inhibition of the AP-1 pathway (SR11302 inhibitor) during the Nrf2−/− thyroid organoid differentiation is not able to reverse the down-regulation in Tg expression (***P = 0.0002 and *P = 0.026; ns = denotes no significant difference between groups). (B) Confocal images of controls (Ctrl WT and Nrf2 KO) and Nrf2 KO organoids (day 22) treated with AP-1 inhibitor, SR11302. Tg (green) and Tg-I (magenta) staining shows that AP-1 inhibition does not reverse the hypothyroid phenotype in Nrf2 KO thyroid organoids. AP-1 inhibition was performed from day 7 to day 22 of the differentiation protocol. Scale bars: 50 μm.
