Schematic showing the experimental approach and procedures in this study. Zebrafish were exposed to each synthetic glucocorticoid from 0 days post fertilisation (dpf) through to 4dpf to assess drug uptake and impacts on embryo-larval development and innate immune system function; and from 0 to 10dpf to determine the effect on jaw cartilage and bone development and density. Inset images illustrate the various methods employed to assess structure and function at each life stage. For embryo-larval development, morphological assessment was employed alongside alcian blue/alizarin red (AB/AR) staining for jaw cartilage and bone (left box). For immune cell function transgenic fish with fluorescently tagged neuromasts were employed to assess cell migration to neomycin-injured neuromasts (circled) (middle box). For jaw cartilage and bone formation and density, transgenic fish with fluorescent cartilage and bone were employed from which jaw structure size and cartilage (red/pink) and bone (green) density were measured (right box).
