Expression of Rab5 Prevents PGCs from Reaching Their Target during Zebrafish Development and Alters the Migratory Shape of PGCs

(A) PGCs, visualized through the expression of GFP:nos1-3′UTR and used as control, correctly migrated to the gonadal area (boxed) by 32 hpf (hours post fertilization). In all control embryos (188/188), PGCs reached the appropriate final gonadal target.

(B and C) In more than 70% of embryos injected with Rab5a:nos1-3′UTR (200/280), a significant number of PGCs were found in ectopic positions throughout the body. Scale bars, 50 μm.

(D) Still images from time-lapse movies showing PGCs coinjected with GFP:nos1-3′UTR and either empty vector (left, GFP, see also Movie S13) or RAB5a:nos1-3′UTR (middle/right, GFP+RAB5, see also Movie S13).
