I am interested in hemopoietic stem cell biology and leukemia. In my previous work, at the EMBL in Heidelberg, we have developed a system of multipotent progenitors transformed by an avian leukemia virus which can be grown in large amounts and can be aeasily handled. Using these cells we showed a role of the transcription factors GATA-1, PU.1, C/EBP a and b in decision making towards eosinophils and myeloblasts. We are now attempting to make transgenic zebrafish in which cells of the myeloid lineage are labeled with a red fluorescent protein. The long term goal is to create bicolored fish through crossing with the GATA-1 EGFP fish from Shuo Lin, that contain green red blood cells. Embryos of these fish will then be used in time lapse experiments, as a testtube for genes that affect lineage decisions (such as the above transcription factors), as recipients for leukemia inducing genes and possibly as a tool to perform a screen for novel genes that play a role in commitment.
