Expression cloning of neural inducing factors in zebrafish: In a cross-species approach zebrafish cDNA libraries are screened for factors with neural inducing activity in a Xenopus animal cap assay. In vitro transcribed RNAs from pools of a zebrafish gastrula stage and adult brain library are injected into early Xenopus embryos. Animal caps are dissected at the blastula stage and cultivated until control embryos reach late neural stages. Induction of neural marker expression in the explants is assayed by RT-PCR with panneural markers as well as regionally restricted neural markers. Subsequent sib selection of positively identified pools with neural-inducing activities will finally lead to the identification of single clones that are involved in the induction and/or patterning of the early vertebrate nervous system. Those factors will be functionally characterized in the available zebrafish mutants with defects in the formation of neural structures.
