Interference with ifi27 gene on cell ROS, apoptosis and mitochondrial membrane potential after GJB2 c.109G > A mutation. After transfecting pcDNA3.1 empty vector, GJB2, GJB2 c.109G > A GJB2 c.109G > A + si-NC, and GJB2 c.109G > A + si-IFI27 into HEK293 cells for 48 h, flow cytometry was used to analyze (A, B) ROS levels and statistical charts, (C, D) cell apoptosis and statistical charts, (E) Fluorescence microscopy was used to observe mitochondrial membrane potential changes in cells subjected to different treatments. n ≥ 3. Each column and bar represent the mean ± SEM. All data were statistically analyzed by one-way ANOVA, * p < 0.05 indicates a significant difference, and *** p < 0.001 indicates an extremely significant difference.
