The nuclear translocation ability of Cas9 protein expressed in HEK293T cells. (A) General structure of transfection constructs. CMV, cytomegaloviral promoter; zCas9, Cas9 with codon optimization for the zebrafish; ERT2, human estrogen receptor with three missense mutations (G400V/M543A/L544A) in the ligand-binding domain; NLS1, SV40 nuclear localization signal; NLS2, nucleoplasmin NLS; pA, polyadenylation signal. (B) Nuclear translocation ability of different Cas9 fusion proteins with or without 4-OHT treatment. The transfected cells were immunostained with Cas9 antibody plus DAPI staining for nuclei 24 hpt.
