dCas9 is a catalytically inactive version of Cas9 that lacks endonucleolytic activity. dCas9 contains two point mutations in both its RuvC-like (D10A) and HNH nuclease (H840A) domains. dCas9 is used in CRISPR interference (CRISPRi) for sequence-specific inhibition of transcription via steric hindrance.
