FUNCTION: Substrate recognition component of a SCF (SKP1-CUL1-F-box protein) E3 ubiquitin-protein ligase complex which mediates the ubiquitination and subsequent proteasomal degradation of target proteins (By similarity). Probably recognizes and binds to phosphorylated target proteins: the interaction with substrates requires the phosphorylation of the two serine residues in the substrates' destruction motif D-S-G-X(2,3,4)-S (By similarity). SCF(FBXW11) mediates the ubiquitination of phosphorylated CTNNB1 and participates in Wnt signaling regulation (By similarity). Participates in Wnt signaling regulation, and plays a role in eye and jaw development (PubMed:31402090). SCF(FBXW11) plays a key role in NF- kappa-B activation by mediating ubiquitination of phosphorylated NFKBIA, leading to its degradation by the proteasome, thereby allowing the associated NF-kappa-B complex to translocate into the nucleus and to activate transcription (By similarity). {ECO:0000250|UniProtKB:Q9UKB1, ECO:0000269|PubMed:31402090}. PATHWAY: Protein modification; protein ubiquitination. {ECO:0000250|UniProtKB:Q9UKB1}. SUBUNIT: Self-associates. Component of the SCF(FBXW11) complex. {ECO:0000250|UniProtKB:Q9UKB1}. SUBCELLULAR LOCATION: Cytoplasm {ECO:0000250|UniProtKB:Q5SRY7}. Nucleus {ECO:0000250|UniProtKB:Q5SRY7}. DEVELOPMENTAL STAGE: At 4 days post fertilization (dpf), it is expressed at low levels in the retina and brain, and at higher levels in the jaw mesenchyme. {ECO:0000269|PubMed:31402090}. DOMAIN: The N-terminal D domain mediates homodimerization. {ECO:0000250|UniProtKB:Q9UKB1}. DISRUPTION PHENOTYPE: Knockdown of both fbxw11a and fbxw11b results in abnormally developed pectoral fins and heart edema by day 3 post- fertilization (3 dpf). At 5 dpf, knocked-down embryos show periocular edema, small and misshapen eyes, and abnormal development of the jaw. {ECO:0000269|PubMed:31402090}. |
