Overexpression of CHD8S in developing zebrafish embryos promotes brain defects. A. Single-cell zebrafish embryos were injected with synthetic chd8s mRNA or egfp mRNA, as a control, both at 150 ng/μl, and observed at 24 hpf. Overexpression produced consistent phenotypes grouped into classes. The chart shows cumulative results (percentages of normal embryos and those with abnormal phenotypes) from injection of 249 embryos with chd8s in three independent experiments, and 58 embryos with egfp. B. Representative examples of normal (wt), or phenotypes are shown. Class 1 embryos show hindbrain ventricle expansion defects (asterisks indicate hindbrain ventricle). Class 2 embryos show hindbrain defects, forebrain deficiencies (arrows indicate optic stalk), and tail abnormalities (arrowheads). Class 3 embryos display cyclopia in addition to hindbrain ventricle and tail abnormalities.
